چکیده

Tumor necrosis factor (TNF)–a inhibitors are a subtype of disease-modifying antirheumatic drugs that have revolutionized the treatment of rheumatologic and gastrointestinal disorders. TNF-a is a cytokine with a complex role in immune system regulation and maintenance of self-tolerance. Despite their clinical benefit in rheumatologic conditions, TNF-a inhibitors have been implicated in the development of CNS and peripheral nervous system disorders. We describe an asymptomatic patient with radiologic and CSF analysis suggestive of CNS demyelination in the setting of TNF-a therapy.

CNS Demyelination with TNF-α Blockers

Tumor necrosis factor-α (TNF-α) blockers are a popular therapeutic choice in a number of inflammatory diseases. Thus far, five TNF- α blockers have been approved for clinical use (etanercept, infliximab, adalimumab, golimumab. and certolizumab). Despite being considered relatively safe, serious side effects associated with immune suppression have been reported, including central and peripheral ...

Effect of Honey Bee Venom on Lewis Rats with Experimental Allergic Encephalomyelitis, a Model for Multiple Sclerosis

Multiple sclerosis (MS) is a progressive and autoimmune neurodegenerative disease of the central nervous system (CNS). This disease is recognized through symptoms like inflammation, demyelination and the destruction of neurological actions. Experimental allergic encephalomyelitis (EAE) is a widely accepted animal model for MS. EAE is created in animals by injecting the tissue of myelin basic...

Pattern of demyelination occurring during anti-TNF-α therapy: a French national survey.

OBJECTIVE To determine the pattern of demyelinating disorders (DDs) occurring during anti-TNF-α therapy. METHODS Between June 2005 and April 2008, 1800 French rheumatologists and internists were contacted to report cases of DDs occurring in patients treated with anti-TNF-α. RESULTS After a median of 10.2 (1.5-39.9) months of treatment, 33 patients developed DDs: 22 had CNS and 11 peripheral...

Matrix metalloproteinase-8 plays a pivotal role in neuroinflammation by modulating TNF-α activation.

Matrix metalloproteinases (MMPs) play important roles in normal brain development and synaptic plasticity, although aberrant expression of MMPs leads to brain damage, including blood-brain barrier disruption, inflammation, demyelination, and neuronal cell death. In this article, we report that MMP-8 is upregulated in LPS-stimulated BV2 microglial cells and primary cultured microglia, and treatm...
